This really depends on how you define the term "first."
 
Generally speaking, we could say that the skin is the first barrier.  However, the linings of the alimentary canal (mouth, esophagus, stomach, etc.) and the linings of the lungs are also called into play, so that we might say that the integumentary system is the first barrier to be more complete in our answer.
